For example, if I sell protein supplements for muscle gain, then keywords for my business would be 'protein supplements', 'protein mass gainers', 'protein powders for muscle gain', and so on.

You can even search for your business on Google and closely study the first page results to see what is the most popular content that's coming up. It'll give you a fair idea of how you should structure your website content, keeping in mind your target audience.
Finally, on identifying the high value keywords, you target them on your website through efforts such as blog writing, using keywords as links, including them in page titles (<h1>, <h2> and <h3>) , meta descriptions, URL structure and image alt descriptions.
